Dihydroberberine inhibits human Ether-related gene (hERG) channels and significantly reduces the expression of heat shock protein 90 (Hsp90) and its interaction with hERG.Dihydroberberine possesses anti-inflammatory, anti-atherosclerotic, hypolipidemic and antitumor activities. Dihydroberberine yellow crystals, soluble in methanol, ethanol, DMSO and other organic solvents, derived from Berberis. UsesDihydroberberine has anti-myocardial ischemia and arrhythmia.
Dihydroberberine (DHB), a hydrogenated derivative of BBR, has been shown to be highly effective in vivo against a number of metabolic diseases and has a favorable safety profile.

Function | DHB exerts its uric acid-lowering effect by inhibiting the activation of XOD, URAT1, GLUT9, and NLRP3 inflammatory vesicles in the kidneys, and is renoprotective in addition to being antihyperuricemic. The treatment of DHB had a protective effect on renal function. These results suggest that DHB ameliorates renal injury and has a nephroprotective effect in HUA mice. DHB showed XOD enzyme inhibition in a concentration-dependent manner.DHB significantly inhibited XOD activity (Figure 4D). This result suggests that DHB has an inhibitory effect on XOD. Treatment with DHB was effective in ameliorating PO/HX-induced renal inflammation. DHB showed the ability to inhibit XOD activity both in vitro and in vivo. And the treatment of DHB also showed significant inhibition of renal URAT1 and GLUT9 levels. Thus, DHB was able to reduce urate formation by inhibiting hepatic XOD while down-regulating renal URAT1 and GLUT9 to enhance urate excretion, suggesting a dual anti-hyperuricemic effect. In addition, the mechanism by which DHB exerts its renoprotective effects was found to be through the inhibition of NLRP3 inflammasome activation to attenuate renal inflammation and dysfunction. |
